Hippo Pathway trans-Reporting System

The transLight Hippo Pathway Luciferase Reporting System is a specialized, in vivo trans-reporter assay designed to provide a rapid and precise read-out of Hippo signaling pathway activity by monitoring the transcription factor TEAD4. This system utilizes a Gal4-TEAD4 fusion protein expressed from the pGAL4-TEAD plasmid, which, in the presence of unphosphorylated YAP, activates luciferase expression from the pHTS-GAL4 reporter plasmid. When the Hippo kinase cascade is activated, YAP becomes phosphorylated and is retained in the cytoplasm, reducing luciferase activity. Conversely, inhibition of Hippo signaling enhances luciferase output, offering a sensitive method to evaluate the pathway's activation status. This system is particularly valuable for studying gene involvement in Hippo signaling, assessing the impact of growth factors, drugs, or extracellular stimuli, and includes well-defined controls and plasmids for accurate interpretation. With high transfection efficiency and adaptability to various mammalian cell lines, transLight enables reproducible, pathway-specific functional studies using luciferase-based quantification.
